PSMD1 Inhibition via LNPs Reduces Myeloma Growth, Overcomes Resistance

Multiple myeloma, a malignancy of plasma cells, presents persistent therapeutic challenges, including acquired drug resistance and relapse following treatment with targeted agents [1]. The proteasome pathway is a central therapeutic axis in this disease, and proteasome inhibitors are a cornerstone of care; however, the emergence of resistance mechanisms often limits their long-term efficacy [2]. Concurrently, advances in drug delivery systems like lipid nanoparticles are proving essential for improving the specificity and reducing the toxicity of therapies, especially for nucleic acid-based agents [3, 4]. There is also growing recognition that modulating immune responses within the tumor microenvironment can enhance therapeutic effect, creating a need for agents that possess both direct anti-cancer and immune-modulatory properties [5, 6]. A recent study investigates a specific vulnerability within the proteasome pathway that may address these unmet needs in multiple myeloma treatment.
