IBD Inflammation Augments CHIP Risk, APE1/Ref-1 Identified as Therapeutic Target

Clonal hematopoiesis of indeterminate potential (CHIP), a condition marked by age-related somatic mutations in hematopoietic stem cells, is an established risk factor for myeloid malignancies, cardiovascular disease, and chronic kidney disease [1, 2, 3]. The condition's pathology is tightly linked to systemic inflammation, which is thought to drive both its progression and its associated comorbidities [4, 5]. Because inflammatory bowel disease (IBD) is a state of profound chronic inflammation, the interplay between these two conditions is of significant clinical interest [1]. Inflammation may promote the expansion of hematopoietic clones, while CHIP itself can amplify a pro-inflammatory state, creating a damaging feedback loop [6, 7]. A recent study now provides specific data on the prevalence of CHIP in IBD and identifies a potential mechanism linking them [1, 8].
