DHODH Inhibition Targets Pyrimidine Synthesis in Chemo-Resistant B-ALL

Relapse remains a primary barrier to long-term survival in B-cell acute lymphoblastic leukemia (B-ALL), with a significant number of patients developing resistance to conventional chemotherapy [1, 2, 3]. This clinical challenge has intensified the search for mechanisms that drive chemo-resistance and for targeted therapeutic strategies that can overcome it [4, 5]. Recent research has increasingly focused on metabolic adaptations, such as altered glucose consumption and nucleotide synthesis, as critical factors in tumor survival and drug resistance [6, 7]. A new study provides a direct link between hyperactive cell signaling pathways and a specific metabolic dependency in these aggressive leukemia cells, revealing a potential therapeutic vulnerability.
